You must install .Net Add-ins for QTP to access with your web based .Net application.

After installation, when you open you QTP, Add-in Manager window will be opened. There you should select Web and .Net Addins and then carry on woth your work.

If you are not able to view Add-in manager, Once you open QTP test script, Goto File menu, Click on Test Settings and Select Properties tab. Click on Modify button and should select Web and .net addins.

After that you are able to prepare your test scripts for .Net application
