How do we fine tune the Mapping and What methods do we follow for different types of Transformations.

When you mean mapping, I assume it to be Informatica Mapping.
Refer to the help in the Informatica Powercenter Designer. Type Performance Tuning and you will get the different methods / techniques that can be used for performance tuning.
Generally performance tuning is done in the following sequence
1. Target bottlenecks - Measure the performance of the mapping by filtering out the records from being written to the target. If there is a change in the time, then the issue is with the target. It could be with the primary key defined.
2. Source bottleneck - Introduce a filter transformation immediately after the source qualifier and filter the records from being passed to the subsequent transformations. If the time taken by the mapping is the same, then the issue is with the source. Generally it happens with Database sources. Try creating indexes or apply appropriate filtering conditons
3. Mapping bottleneck- It could be because of various transformations within the mapping. Generally because of Joiner, Sorter, aggregator. Transformations that uses Cache. Try and give appropriate cache sizes
4. Last in case if you are sequence generator transformation, check the value in the number of cached cycles. It should not be 1. In general a value of 1000 is preferred.
